Built On Decades of Science
CV6 Therapeutics was built around a long-term scientific pursuit: understanding how widely used cancer therapies may be therapeutically enhanced.
Led by Dr. Robert Ladner and supported by a team of scientists, clinicians, and advisors, the company brings together decades of research focused on nucleotide metabolism and treatment resistance. That work has evolved into a clinical-stage program with potential relevance across multiple cancers, with additional inflammatory disease programs under development.
The Question That Started It
For more than three decades, Dr. Robert Ladner has focused on a fundamental question in cancer biology that others largely overlooked: how the mismanagement of uracil in DNA influences the activity of widely used therapies such as 5-fluorouracil, and how this biology contributes to treatment resistance.
A Different Path in Oncology
While much of the field moved toward entirely new drug classes, his work remained anchored in a different belief: that understanding and targeting this mechanism could meaningfully extend the therapeutic utility of therapies already used worldwide.
Building the CV6 Research Plan
That long-term commitment led to the development of the CV6 Therapeutics approach, centered on selectively targeting dUTPase and disrupting nucleotide metabolism in cancer cells. By increasing uracil incorporation into DNA, CV6-168 is designed to modulate the activity of 5-FU and is being investigated in settings associated with treatment resistance.
From Hypothesis to Clinic
Today, that vision is advancing in the clinic, supported by decades of research and a growing body of translational and clinical evidence.

That mindset has defined Dr. Ladner’s career. While much of the industry moved toward entirely new therapies, he remained focused on understanding how established cancer treatments could be made more effective.
